A UCLA-led mouse study found that a single rapamycin dose rapidly improved autism-like symptoms in adults exposed to prenatal inflammation, reversing brain overactivity, seizures, and repetitive behaviors within hours. However, effects are temporary, and repeated dosing loses efficacy, making rapamycin unsuitable for human treatment. Researchers suggest the findings highlight potential targets for future therapies focusing on brain functioning rather than structural changes.

Pregnant mice exposed to mild inflammation had offspring with lasting autism-like traits, including sensory sensitivity and abnormal brain networks. When adult offspring received rapamycin, symptoms improved rapidly, though temporary. The drug’s effects stem from rebalancing neuronal excitability and gene expression tied to autism and epilepsy, not structural brain repair.

Co-author Dr. Neil Harris emphasized the temporary nature of rapamycin’s benefits and its toxicity risks. Study funding came from the Adelson Medical Research Foundation, NIH, Simons Foundation, and UCLA’s autism research centers.
